Objectives:To assess the effectiveness and safety of oral sildenafil, a new oral phosphodiesterase 5 inhibitor, in patients with erectile dysfunction (ED).

Methods:A prospective, randomized, parallel-group, double-blind, placebo-controlled, multicenter, double-masked, fixed-dose, 24-h crossover study was conducted on 23 patients with mild to moderate ED secondary to erectile dysfunction, who were randomized to receive a single oral sildenafil, or placebo. Patients were assessed at baseline and 1, 2, 4, 6, and 8 weeks by a physician, and then, after 8 weeks, at weeks 0, 1, 2, 4, 6, and 8 weeks, by a physician and a patient nurse, the physician assessed the frequency of sexual activity and any adverse events related to sexual activity. The frequency of sexual activity was assessed by a physician. Sexual activity was assessed by the physician, the patient nurse, and a physician-patient relationship. The safety of sildenafil was assessed by a physician, the patient nurse, and a physician-patient relationship.

Results:Of 23 patients randomized, 14 (96%) were on the placebo and 13 (90%) were on sildenafil. The frequency of any adverse events related to sexual activity was significantly lower in the sildenafil group (6.7% vs. 8.2%) and significantly lower in the placebo group (14.5% vs. 3.9%). The safety of the placebo was assessed by the physician, the patient nurse, and a physician-patient relationship.

Conclusions:The efficacy and safety of oral sildenafil in patients with ED are satisfactory.

Sildenafil is an oral phosphodiesterase 5 inhibitor (PDE5I). Efficacy and safety were evaluated using two randomized, double-blind, placebo-controlled studies in adult men with erectile dysfunction. Sildenafil showed superiority in the efficacy and safety of the oral PDE5I in patients with ED.

Introduction

Erectile dysfunction (ED) is one of the most common sexual problems in men. The global prevalence of ED has increased with increasing age.
